Target Information
The munc-18 interacting protein (Mint) family consists of evolutionarily conserved adaptor proteins involved in membrane transport and organization. In mammals, three isoforms exist: Mint1, Mint2, and Mint3.
- The C-terminal half of these proteins is highly conserved and contains PTB and PDZ domains that mediate protein interactions.
- Mint1 and Mint2 are brain-specific and bind to Munc18, a key component of synaptic vesicle fusion.
- Mint3 is ubiquitously expressed, at the lowest levels in brain and testis, and does not interact with Munc18.
- Mint3 interacts with the Alzheimer’s disease-related amyloid precursor protein (APP) via its PTB and PDZ domains, potentially linking APP to transport machinery components that regulate its trafficking, endocytosis, and metabolism.
- Abnormal APP metabolism is implicated in early-onset Alzheimer’s disease.
