Target Information
Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase family, which includes EGFR, ERBB2, and ERBB3. ERBB4 is activated by neuregulins and other ligands, inducing cellular responses such as mitogenesis and differentiation. It mediates tyrosine phosphorylation and plays a key role in signal transduction processes including metabolism regulation, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
ErbB4 is highly expressed in several breast carcinoma cell lines (T47-D, MDA-MB-453, BT-474, H3396) and in normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum. Expression is lower or absent in some breast tumor lines (MDA-MB-231, MDA-MB-157, MDA-MB-468, SKBR-3). Elevated ErbB4 levels in certain tumor cell lines suggest a potential role in malignancy.
