This product is diluted and ready-to-use.
Recommended positive control tissue: Tonsil (other tissues may also be used).

Designed for professional laboratory use in detecting p40 (DNp63) in formalin-fixed, paraffin-embedded tissues by manual qualitative IHC.
Used after primary tumor diagnosis via conventional histopathology.

p63 has two major isoforms: TAp63 and DNp63, differing in their N-terminal domains.

• TAp63 (recognized by anti-p63 antibody): contains a transactivation domain homologous to p53, regulating growth-inhibitory genes.
• DNp63 (recognized by anti-p40 antibody): contains a transcriptionally inactive domain antagonizing TAp63 and p53 activity.

The p40 (clone ZR8) antibody recognizes DNp63 exclusively, serving as a squamous cell carcinoma–specific marker.
It reacts with most squamous cell carcinomas but not with adenocarcinomas, aiding differentiation of lung squamous cell carcinoma vs. poorly differentiated adenocarcinoma.
Also applicable as a basal cell/myoepithelial marker for distinguishing prostate adenocarcinoma vs. benign glands and breast intraductal vs. invasive ductal carcinoma.

Recommended Usage

• Use with formalin-fixed, paraffin-embedded sections.
• Pretreat tissue with heat-induced or enzymatic epitope retrieval.
• Visualize antigens via sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate.
• Interpret results under a light microscope.

Quality Control

• Run a positive tissue control with every staining procedure.
• Positive control tissues should contain both positive and negative staining components.
• Use fresh, properly fixed and processed specimens for external positive controls.
• Weakly positive tissues are preferred to detect minor reagent degradation.
• Include internal or external negative controls to verify absence of nonspecific staining.

Target Information

The encoded protein functions as a cytokine regulating hematopoietic cell proliferation and apoptosis via the IL9R receptor, activating STAT pathways. It is a candidate gene for asthma and plays a role in bronchial hyperresponsiveness.
