Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells throughout development except in terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with BCR to induce tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and tested for flow cytometry using normal human peripheral blood cells. Use 5 µL (0.4 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Use 5 µL of CellBlox Plus per sample or per 100 µL of cell sample for single-color controls.

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence with minimized spillover and spread.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and certain myeloid leukemia cells. CD19 serves as a signal-amplifying coreceptor for the B cell receptor (BCR), regulating B cell activation and proliferation. Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
