Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Murine TNF-alpha. Anti-Murine TNF-alpha-specific antibody was purified by affinity chromatography employing an immobilized Murine TNF-alpha matrix.
Sandwich ELISA:
To detect Murine TNF-alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Murine TNF-alpha (500-P64Bt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Murine TNF-alpha.
Western Blot:
To detect Murine TNF-alpha by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Murine TNF-alpha is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers of two to five units and shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest a neuroprotective function, and TNF alpha causes tumor necrosis when injected into tumor-bearing mice. It also plays a role in immune responses to bacterial, viral, parasitic, and fungal infections, and has been implicated in autoimmune diseases, insulin resistance, and cancer.
