Target Information
Ago proteins are broadly expressed in somatic cells and associate with miRNAs, playing key roles in RNA silencing pathways. Among them, Ago2 uniquely exhibits endonucleolytic “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when base-pairing is perfect. With partial complementarity, Ago2 mediates translational repression instead of cleavage.
Ago2 is a core component of the RISC complex, initiating degradation of target mRNAs through siRNA or miRNA-guided gene silencing. It also functions independently of Dicer in RNA slicing and regulates miRNA maturation. Overexpression of Ago2 has been linked to tumor cell growth and cancer progression. Mouse studies confirm its essential role in embryonic development and regulation of B-lymphoid and erythroid development.
