Target Information

The KCNK16 gene encodes the TALK-1 channel, an abundant K⁺ channel transcript found in mouse and human beta-cells.
There are four human TALK-1 transcript variants, including two functional forms (TALK-1a and TALK-1b).
The TALK-1 structure includes four transmembrane domains, two extracellular loops, and intracellular N- and C-termini.

  • The C-terminal tail influences the magnitude of channel activation.
  • The N-terminus is important for protein interactions.

TALK-1 (two-pore domain K⁺ channel, K2p 16.1) modulates glucose-stimulated insulin secretion dependent on beta-cell Ca²⁺ influx.
It produces outwardly rectifying, non-inactivating K⁺ currents enhanced by elevated extracellular pH.
The channel is pH-sensitive and activated by singlet oxygen and nitric oxide.
