Target Information

FOXK1 is a transcriptional regulator involved in various processes such as glucose metabolism, aerobic glycolysis, muscle cell differentiation, and autophagy. It recognizes and binds the forkhead DNA sequence motif (5'-GTAAACA-3') and can act as a transcription activator or repressor depending on the context.
Together with FOXK2, FOXK1 regulates metabolic reprogramming toward aerobic glycolysis, promoting glycolytic enzyme expression (e.g., HK2, PFK, PKLR, LDH) while suppressing pyruvate oxidation via up-regulation of PDK1 and PDK4.
It plays a role in gluconeogenesis during fasting and participates in mTORC1-mediated metabolic reprogramming by regulating glycolysis-related genes such as HIF1A.
FOXK1 also acts as a negative regulator of autophagy in skeletal muscle, repressing autophagy gene expression during starvation.
Additionally, it regulates muscle progenitor cell proliferation by repressing FOXO4 activity and contributes to muscle repair and remodeling.
FOXK1 positively regulates Wnt/β-catenin signaling and may reduce virus replication by promoting antiviral gene expression.
