This antibody is provided in a ready-to-use formulation.
Recommended positive control tissue: Kidney (not limited to this tissue type).
Intended for use by laboratory professionals in detecting the target protein in formalin-fixed, paraffin-embedded tissues via manual qualitative IHC testing.

Galectin-3 belongs to the β-galactosidase-binding lectin family and is involved in cell growth, adhesion, inflammation, mRNA processing, and apoptosis. Its aberrant expression is associated with malignant transformation and metastasis in breast, colon, and thyroid carcinomas. Galectin-3 reactivity is observed in neutrophil nuclei, vascular endothelium, and carcinoma tissues. It is useful for differentiating benign and malignant thyroid neoplasms and identifying certain liver disorders.

For optimal staining:

• Use formalin-fixed, paraffin-embedded sections.
• Apply heat-induced or enzymatic epitope retrieval before staining.

IHC staining visualizes antigens through sequential application of:

1. Primary antibody
2. Secondary antibody
3. Enzyme complex
4. Chromogenic substrate

The resulting visible reaction product is interpreted under a light microscope to aid differential diagnosis.

Controls

• Positive Control: Run with every staining procedure; should include both positive and negative staining components.
• External Positive Control: Fresh, well-characterized specimens with weak positive staining ensure quality control and detect subtle reagent degradation.
• Negative Control: May be internal or external; should show absence of specific staining to confirm validity of results.

Target Information

Galectin-3 is a galactose-specific lectin that binds IgE and mediates endothelial cell migration via α3β1 integrin and CSPG4. It cooperates with DMBT1 in epithelial cell differentiation during embryogenesis and acts as a pre-mRNA splicing factor in the nucleus. Involved in acute inflammatory responses, including neutrophil activation, monocyte/macrophage chemoattraction, opsonization of apoptotic neutrophils, and mast cell activation.
