Product Specific Information

The 13A4 monoclonal antibody recognizes mouse Prominin-1 (CD133), a 115–120 kDa pentaspan transmembrane glycoprotein expressed on primitive cells such as hematopoietic, neural, and endothelial stem cells, retina, and developing epithelium. The 13A4 antibody does not cross-react with rat, human, chicken, or Drosophila antigen but may react with canine antigen.

Applications Tested: Tested on mouse bone marrow cells at ≤1.0 µg/test (100 µL final volume). Optimize titration for best performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by a 405 nm violet laser and emits at 600 nm. Use a 610/20 bandpass filter. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact.

Target Information

CD133 (AC133) is a 97 kDa glycoprotein with five transmembrane domains, appearing as a 120 kDa band due to glycosylation. It is a cell surface marker of immature hematopoietic stem cells and other progenitor cells, including neural and embryonic stem cells. CD133 is also expressed in some cancers (leukemias, brain tumors) and at low levels in kidney, pancreas, placenta, and fetal liver. Mutations can cause retinitis pigmentosa and Stargardt disease. Expression varies by tissue via multiple promoters and transcript variants.
