Target Information
RAP80 was initially identified as a zinc-finger containing nuclear protein highly expressed in testis and interacting with the retinoid-related testis-associated receptor (RTR). Subsequent studies showed that RAP80 is recruited by the Coiled-coil domain 98 (CCDC98) protein to BRCA1, enabling BRCA1 foci formation in response to DNA damage caused by ionizing radiation. Both RAP80 and CCDC98 are essential for DNA damage resistance, G2-M checkpoint control, and DNA repair. Cells depleted of RAP80 or CCDC98 exhibit increased sensitivity to ionizing radiation, suggesting that these proteins mediate part of BRCA1’s DNA damage response. At least four isoforms of RAP80 are known to exist.
