Product Specific Information

The YN1/1.7.4 monoclonal antibody reacts with mouse CD54 (ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed by monocytes, lymphocytes, dendritic cells, and endothelial cells. Expression of CD54 is upregulated on activated lymphocytes. Interaction with ligands CD11a and CD11b is important in the inflammatory response. The YN1/1.7.4 clone recognizes a different epitope than the eBioKAT-1 monoclonal antibody.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.4 µg/test (100 µL final volume). Cell count may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Each product contains:

  • 1 vial of CellBlox Plus Blocking Buffer

NovaFluor dyes compatibility:
Not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Improved formulation over CellBlox. Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails prior to combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with reduced spillover and spread impacts.

Target Information

ICAM-1 (CD54) is an 85–110 kDa type I membrane glycoprotein with five immunoglobulin-like extracellular domains, a transmembrane region, and a cytoplasmic domain. It has seven potential N-linked glycosylation sites and shares sequence homology with ICAM-2 and ICAM-3.

ICAM-1 binds integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for Rhinovirus. It is expressed by activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, B and T lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils. Expression increases upon inflammation. Soluble ICAM-1 is detectable in plasma and elevated in inflammatory conditions.
