This product is supplied in a ready-to-use formulation.
A recommended positive control tissue is the adrenal gland, although other positive tissues may be used.

The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded (FFPE) tissue via manual qualitative immunohistochemistry (IHC). It should be used after the primary diagnosis has been made using conventional histopathology.

Steroidogenic Factor 1 (SF-1) is a transcription factor that regulates genes involved in the development and function of adrenal and reproductive glands. Encoded by the NR5A1 gene on chromosome 9q33.3, SF-1 is expressed in the adrenal cortex, adrenal cortical adenomas, carcinomas, and sex-cord stromal tumors of testicular and ovarian origin.

For optimal staining, use heat-induced or enzymatic epitope retrieval on deparaffinized tissue.
IHC staining visualizes antigens through sequential application of:

  1. Primary antibody (specific to antigen)
  2. Secondary antibody (link antibody)
  3. Enzyme complex
  4. Chromogenic substrate

The enzymatic reaction produces a visible product at the antigen site, which is interpreted microscopically.

A positive tissue control must be included in every staining run. This control should contain both positive and negative staining areas. External positive controls should be prepared similarly to patient samples and ideally exhibit weak positive staining to detect subtle changes in antibody sensitivity. Such controls ensure reagent stability and staining consistency.

Negative control tissues (internal or external) are used to confirm specificity and assess background staining. If specific staining appears in negative controls, results should be considered invalid.


Target Information

Steroidogenic Factor-1 (SF-1) is an orphan nuclear receptor essential for the development and differentiation of endocrine organs including the hypothalamus, pituitary, gonads, and adrenal glands.
SF-1 regulates the transcription of multiple steroidogenic enzymes (e.g., cytochrome P450 family members) and contains two activation domains (AF1, AF2) necessary for interactions with coactivators and corepressors such as SRC-1, GRIP1, CBP, and SMRT.
