Target Information
Petosemtamab is a biosimilar that targets EGFR.
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans.
EGFR belongs to the HER/ERbB family of proteins that includes ERbB2, ERbB3, and ERbB4. It is a transmembrane receptor; binding of ligands such as EGF and TGF-α to its extracellular domain induces dimerization and autophosphorylation of tyrosine residues in the cytoplasmic domain.
EGFR activation triggers multiple signaling cascades (Ras-ERK, PI3K-Akt, Jak-STAT, PKC) that regulate cell growth and proliferation.
Phosphorylation of EGFR at Y1086 allows GRB2 binding and MAPK pathway activation.
Activated EGFR undergoes endocytosis and is either degraded or recycled.
Mutations in EGFR are associated with lung cancer, and overexpression correlates with poor prognosis in various tumors, including those of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, and esophagus.
