Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily that includes EGFR, ERBB2, and ERBB3.
ERBB4 binds and is activated by neuregulins and other factors, inducing various cellular responses such as mitogenesis and differentiation.
ErbB4 mediates tyrosine phosphorylation upon activation by heregulin and regulates essential cellular processes including metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
It is predominantly expressed in breast carcinoma cell lines and normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum.
Expression levels vary among breast tumor cell lines, being highest in T47-D, MDA-MB-453, BT-474, and H3396, and lower or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3.
Elevated ERBB4 levels in certain tumor cell lines suggest its potential role in oncogenesis.
