Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was first identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30+ lymphomas, acting as a costimulatory molecule in thymic negative selection. It is expressed on Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (e.g., Burkitt’s), virus-infected T/B cells, and activated normal T/B cells.

In T cells, CD30 expression is found on subsets producing Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. Soluble CD30 (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30, mediating NF-κB activation. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, contributing to autoimmunity protection.

Two alternatively spliced transcript variants encode distinct isoforms. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
