Invitrogen™ Dynabeads™ kilobaseBINDER™ Kit

The Dynabeads kilobaseBINDER Kit is designed for immobilizing linear, biotinylated, long double-stranded (ds) DNA molecules (>2 kb). It combines streptavidin-coupled Dynabeads and a unique binding solution containing a patented immobilization activator that significantly increases the binding capacity for linear, large dsDNA fragments. This product provides an advantage whenever immobilization of a large quantity of long, biotinylated dsDNA is required.

Consistency and Reproducibility

The monolayer of streptavidin leaves most biotin-binding sites available for free biotin and biotinylated ligands. The defined surface enables efficient capture, separation, and downstream handling. Low bead size distribution (<5%) and streptavidin monolayer ensure negligible leakage, providing batch consistency and reproducibility.

Binding Capacity

The ligand size and biotinylation procedure affect binding capacity, which also depends on steric availability and charge interactions. The Dynabeads kilobaseBINDER Kit activates long, linear dsDNA to neutralize steric hindrance, facilitating efficient binding to the bead surface.

Typical binding capacity per mg beads:

• ~70 pmoles of 4 kb biotinylated DNA fragment
• ~80 pmoles of 10 kb fragment

• 195 μg (6 pmol) biotinylated 50 kb DNA fragment

The binding solution increases the binding capacity of biotinylated 4 kb DNA fragments more than 10-fold compared to not using the solution.

Applications

Useful for applications in cell biology such as:

• Building chromatin beads that mimic chromosomes
• Constructing synthetic nuclei for studying RNA export in vitro
• Reconstituting nuclear movement along microtubules
• Target enrichment for long-read sequencing

For long-read sequencing, it is recommended to extend the biotinylated probe by PCR after hybridization to ensure double-stranded DNA formation.

The 2.8 μm magnetic beads are ideal for high-throughput enrichment and can be automated using KingFisher Sample Purification Systems or liquid handlers.
