Target Information

Mitochondrial transcription factor A (TFAM) is a nuclear-encoded protein that translocates to mitochondria, where it plays a vital role in maintaining and expressing mitochondrial DNA (mtDNA). It regulates mtDNA replication and transcription, ensuring proper mitochondrial function. TFAM contains an LC3 interacting region (LIR) motif, enabling interaction with autophagy proteins like LC3, which supports the autolysosomal pathway to remove leaked mtDNA and reduce inflammation.

TFAM expression levels vary among tissues and influence mitochondrial performance. Elevated TFAM can repress mtDNA expression in skeletal muscle, contributing to oxidative phosphorylation (OXPHOS) deficiencies, while in cardiac tissue, increased mtDNA copy number helps maintain OXPHOS balance. TFAM is essential for mtDNA packaging, stability, and replication, and its dysregulation is linked to neurodegenerative and mitochondrial diseases.
