Target Information

ARC (also known as NOL3 - Nucleolar protein 3) has two isoforms:

• Isoform 1: Involved in RNA splicing.
• Isoform 2: Functions as an apoptosis repressor blocking multiple modes of cell death.
  ARC inhibits extrinsic apoptotic pathways by:

1. Interacting with FAS and FADD to block DISC assembly upon FAS activation.
2. Interacting with CASP8 in a mitochondria localization- and phosphorylation-dependent manner, limiting soluble CASP8 for DISC-mediated activation.

ARC also inhibits intrinsic apoptotic pathways through interactions with BAX, resulting in BAX inactivation and preventing mitochondrial dysfunction and release of pro-apoptotic factors.
Mutations affecting the gene can result in myoclonus, familial cortical disorders.
