Target Information

FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It functions as a coactivator or corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region. Association with PAX3 is implicated in alveolar rhabdomyosarcoma. FOXO1 is linked to anti-tumor activity, autophagy, and myogenic differentiation. It is a key target of insulin signaling, regulating metabolic homeostasis under oxidative stress. FOXO1 binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE), and regulates redox balance, osteoblast numbers, and bone mass. It orchestrates skeletal endocrine function, suppresses RUNX2 activity, and promotes gluconeogenesis in hepatocytes by activating genes such as IGFBP1, G6PC, and PCK1. FOXO1 acts downstream of CDK1, PKB/AKT1, and STK4/MST1 to regulate cell death.
