Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. It also exists in a soluble form (~26 kDa). Stimulation of FAS leads to aggregation of intracellular FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases, triggering apoptosis. FAS has been shown to activate NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and participate in proliferative signaling in fibroblasts and T cells. Multiple alternatively spliced transcript variants exist, with some isoforms lacking the transmembrane domain, negatively regulating apoptosis. The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
