Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets. It possesses tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL of cell suspension for single-color controls.
• NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, accounting for ~10% of lymphocyte surface proteins. It regulates Src-family kinases, is essential for T- and B-cell receptor signaling, and modulates cytokine receptor pathways. CD45 isoforms arise from alternative splicing and differ in extracellular domains, influencing membrane localization and cell-type specificity. It also impacts cell survival, apoptosis, and integrin-mediated signaling.
