Target Information
Crystallins are separated into two classes: taxon-specific (enzyme type) and ubiquitous. The ubiquitous class constitutes the major proteins of vertebrate eye lens, maintaining transparency and refractive index. Lens fiber cells lose their nuclei during development, so crystallins remain throughout life, making them extremely stable. Mammalian crystallins are divided into alpha, beta, and gamma families. Beta and gamma crystallins form a superfamily, with alpha and beta further subdivided into acidic and basic groups. Crystallins contain seven regions: four homologous motifs, a connecting peptide, and N- and C-terminal extensions. Gamma-crystallins are monomeric, highly symmetrical proteins lacking connecting peptides and terminal extensions, and are differentially regulated after early development. The encoded protein was initially considered a beta-crystallin but shows greater similarity to gamma-crystallins, being the most significant gamma-crystallin in adult lens tissue. Aging or mutations in these genes are associated with cataract formation.
