Product Specific Information

The 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin of the Ig superfamily that binds sialic acid-containing glycoconjugates. Siglec F is primarily expressed on eosinophils and alveolar macrophages, with lower expression in immature myelomonocytic cells. It is a functional ortholog of human Siglec 8 but not expressed on mouse mast cells.

• Recognizes a formaldehyde-fixed epitope.
• Reported for use in flow cytometric analysis.
• Tested by flow cytometry of thioglycolate-elicited peritoneal macrophages.
• Recommended usage: ≤0.125 µg/test in 100 µL final volume (10⁵–10⁸ cells/test).
• Optimize antibody titration for best performance.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Staining buffer recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic acid-dependent binding. It binds both α2,3- and α2,6-linked sialic acids, with potential masking by cis interactions. Expressed in monocytic/myeloid lineage cells, SIGLEC5 is abundant in leukocytes, spleen, and bone marrow, with lower levels in lymph node, lung, and other tissues. Expressed by monocytes and neutrophils, absent in early leukemic cell lines.
