Target Information
Sonic Hedgehog (SHH) is expressed only during embryogenesis and plays a crucial role in early embryonic patterning. It is involved in ventral neural tube formation, anterior-posterior limb axis development, and ventral somite patterning.
The protein is synthesized as a precursor that autocatalytically cleaves into two parts:
- N-terminal portion: Soluble and responsible for signaling activity
- C-terminal portion: Involved in precursor processing and attaches a cholesterol moiety to the N-terminal product, restricting its diffusion.
Defects in SHH or its signaling pathway can cause holoprosencephaly (HPE), characterized by forebrain malformation and facial deformities. Mutations may also contribute to VACTERL syndrome, involving vertebral, anal, tracheoesophageal, renal, cardiac, and limb abnormalities.
