Target Information

Piezo1 (FAM38A) is a pore-forming subunit of a mechanosensitive non-specific cation channel that generates currents characterized by a linear current-voltage relationship sensitive to ruthenium red and gadolinium. Structurally, Piezo1 contains 36 transmembrane domains and functions as a homotetramer.

Piezo1 plays a key role in epithelial cell adhesion by maintaining integrin activation through R-Ras recruitment to the ER and subsequent stimulation of calpain signaling. In the kidney, Piezo1 contributes to the detection of intraluminal pressure changes and urine flow sensing. It acts as a shear-stress sensor that promotes endothelial cell organization and alignment in the direction of blood flow through calpain activation, playing a key role in blood vessel formation and vascular structure during development and adult physiology.

Defects in the PIEZO1 gene are associated with dehydrated hereditary stomatocytosis, pseudohyperkalemia, perinatal edema, and lymphedema.
