Target Information
Paraoxonase-1 (PON1) is a calcium-dependent ester hydrolase associated with apoA-I in HDL. It prevents oxidation of LDL, reducing formation of atherogenic oxidised-LDL. Human PON1 polymorphisms are linked to coronary artery disease, and reduced PON1 activity is observed in individuals predisposed to atherosclerosis, such as those with diabetes, familial hypercholesterolemia, or renal disease.
PON2 is an intracellular protein protecting cells from oxidative damage, while PON3 has similar activity to PON1 but different substrate specificity. Both PON1 and PON3 are implicated in lowering the risk of coronary artery disease and atherosclerosis. Human PON3 shares three conserved cysteine residues with PON1, important for in vivo activity. PON1-based catalytic scavengers may be useful for organophosphate detoxification.
