Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor belonging to the EGF-TM7 family. It contains seven EGF-like extracellular domains, seven transmembrane regions, and a cytoplasmic domain homologous to TM7 superfamily members.

F4/80 antigen is expressed by most mature macrophages and is a key marker for this population. It can also be found on peritoneal eosinophils, Langerhans cells, and certain dendritic cell subtypes. Expression begins early in myeloid development and can be upregulated by M-CSF stimulation. Some macrophage subsets, especially in lymphoid tissues, may lack F4/80 expression.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse peritoneal exudate cells. Recommended ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best results.

eFluor® 660 replaces Alexa Fluor® 647. Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.
Ensure instrument compatibility for detection.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein, member of the EGF-TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies during macrophage maturation and activation. Found in diverse macrophage populations including Kupffer cells, Langerhans cells, microglia, and macrophages in spleen, thymus, and bone marrow stroma.

Absent in macrophages of T-cell areas of spleen and lymph nodes. Potential role suggested in generation of efferent CD8⁺ regulatory T cells.
