Target Information

NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and p53 regulation.
NPM1 shuttles between nucleus, cytoplasm, and nucleolus, chaperoning core histones and preventing protein aggregation. It aids enzyme protection during thermal denaturation and assists renaturation of denatured proteins.
NPM1 is associated with centrosomes and acts as a substrate for CDK2/cyclin E during centrosome duplication. It is linked to tumor-associated translocations forming fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, serving as an interface for oligomerization and oncogenic conversion.
Additionally, NPM1 sequesters tumor suppressor RF in the nucleolus to prevent degradation. Dysfunction of NPM1 is associated with acute myeloid leukemia and lymphomatoid papulosis.
