Product Specific Information

Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature. Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.


Target Information

E-Cadherin (epithelial cadherin) is a classical cadherin from the calcium-dependent adhesion protein superfamily. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. Mutations in this gene are correlated with gastric, breast, colorectal, thyroid, and ovarian cancer. Loss of function is thought to contribute to cancer progression by increasing proliferation, invasion, and/or metastasis. The ectodomain mediates bacterial adhesion to mammalian cells, and the cytoplasmic domain is required for internalization. Identified transcript variants arise from mutations at consensus splice sites. E-cadherin plays a central role in tissue architecture and integrity. In humans, E-cadherin is encoded by the CDH1 gene on chromosome 16. Reduction or loss of E-cadherin expression in carcinomas correlates positively with tumor invasion and metastasis.
