작동 원리

GTPγS 처리로 Rac1을 GTP-bound 활성형으로, GDP 처리로 비활성형으로 전환하여 비교 검출합니다.
활성형 Rac1은 강한 신호를, 비활성형은 미약하거나 신호가 없습니다.
Western blot 검출은 HRP-conjugated secondary antibody (Goat Anti-mouse IgG, Part No. 31430) 및 SuperSignal West Pico Chemiluminescent Substrate (Part No. 34080)를 사용합니다.
30회 pull-down assay 수행이 가능합니다.

Rac1 배경 정보

Rho family GTPases는 세포 신호전달, 전사 조절, actin cytoskeleton 조직화 등 다양한 기능을 수행합니다.
이들은 Rho (RhoA, RhoB, RhoC), Rac (Rac1, Rac2, Rac3, RhoG), Cdc42, Rnd, RhoBTB, Miro family로 구성됩니다.
Rac1 활성화는 actin 중합을 유도하여 세포 주변부에서 membrane ruffling 및 lamellipodia 형성을 유발합니다.
Rac1은 성장인자(EGF, insulin, PDGF, NGF), integrin(fibronectin), chemoattractant(fMLP)에 의해 자극되며, PI3 kinase 및 MAPK 경로(JNK, p38)를 활성화합니다.
Pak1과 phosphoinositol 4-phosphate 5-kinase는 Rac1의 주요 effector로서 각각 JNK 경로 활성화 및 actin filament 조립을 촉진합니다.
