Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it serves as a ligand for PD-1, an inhibitory immune checkpoint molecule. PD-L2 is primarily expressed in dendritic cells but can be induced in various cell types depending on the microenvironment. Its expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. PD-L2 binds PD-1 with higher affinity than PD-L1 and plays a role in regulating immune responses by inhibiting T cell activation and cytokine secretion. Additionally, PD-L2 interacts with RGMb, influencing respiratory tolerance through BMP and neogenin signaling pathways. The human PD-L2 monomer comprises a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
