Target Information
DOK-6 (Docking Protein 6)은 p62 Dok 패밀리의 Dok-4/5 하위 그룹에 속하는 새로운 세포내 어댑터 단백질입니다.
이 단백질은 발달 중인 중추신경계에서 높게 발현되며, Ret 수용체와 함께 교감, 감각, 부교감 신경절 및 신장 발생 중 요관 싹 등 여러 부위에서 공동 발현됩니다.
Pull-down assay 결과, Dok-6의 PTB 도메인은 GDNF로 활성화된 Ret의 인산화된 Tyr(1062) 잔기와 결합함이 확인되었습니다. 또한 Ret의 리간드 활성화는 Dok-6의 C-말단 영역 내 티로신 잔기의 인산화를 유도하며, 이는 주로 Src 의존적 메커니즘을 통해 일어납니다.
따라서 Dok-6는 Ret-Src 신호전달 경로의 기질로 작용합니다.
