Product Specific Information

  • This product is diluted and in a ready-to-use formulation.
  • Recommended positive control tissue: Uterus (not limited to this type).
  • Intended for laboratory professional use in detection of vimentin in formalin-fixed, paraffin-embedded tissue stained in manual qualitative IHC testing.
  • Reacts with a 58 kDa vimentin protein; no cross-reaction with desmin, keratin, neurofilament, or glial fibrillary acid protein.
  • Useful in tumor sub-classification when used in panels with other antibodies.
  • Expression of vimentin with anti-keratin aids in distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas.
  • Labels mesenchymal cells such as melanocytes, lymphocytes, endothelial cells, and fibroblasts.
  • Non-reactivity can assist in identifying tumors lacking vimentin (e.g., hepatoma, seminoma).
  • Suitable as a tissue process control reagent.

Usage Notes

  • Use with formalin-fixed, paraffin-embedded sections.
  • Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
  • IHC staining involves sequential application of primary antibody, link antibody, enzyme complex, and chromogenic substrate, followed by washing steps.
  • Positive tissue control must be run with every staining procedure.
  • External positive control materials should be processed identically to patient samples and selected for weak positive staining to ensure reagent sensitivity.
  • Internal or external negative controls should be used to confirm absence of non-specific staining.

Target Information

Vimentin is a developmentally regulated intermediate filament protein (IFP) found in cells of mesenchymal origin.
It is involved in intracellular transport between the nucleus and plasma membrane and co-expressed with desmin during early development.
Vimentin participates in steroid synthesis regulation by serving as a storage network for cholesterol-containing lipid droplets.
Phosphorylation by protein kinase triggers filament breakdown and cytoskeletal changes that enhance cholesterol transport to mitochondria, increasing steroid synthesis.
