Target Information

Interleukin-18 binding protein (IL-18BP) is a constitutively secreted glycoprotein acting as a natural antagonist for interleukin-18 (IL-18). It prevents IL-18 from binding to its receptors, thereby neutralizing its proinflammatory role in cell-mediated immune responses. This results in reduced IFN-gamma production by T helper type I cells and inhibition of CTL activity through enhanced NK cell cytotoxicity.
IL-18BP is essential for skin homeostasis and has been associated with diseases such as eczema, asthma, and Crohn’s disease. Upregulation by keratinocytes expressing HPV E7 oncoprotein decreases IL-18-induced IFN-gamma production and T-cell activation. Elevated IL-18 and IL-18BP levels, along with proinflammatory cytokines (IFN-gamma, TNF-alpha, IL-1beta, IL-8), are linked to chronic inflammation and observed in macrophages from Crohn’s disease lesions.
IL-18BP belongs to the immunoglobulin-like receptor class and contains a single Ig domain; two isoforms (b and d) have incomplete domains, reducing binding affinity.
