Invitrogen™ BacMam Histone H3K4me3 Cellular Assay Kit

The BacMam Histone H3K4me3 Cellular Assay is a high-throughput screening (HTS) compatible TR-FRET assay designed to interrogate trimethylation at lysine 4 on histone H3 in a cellular format. Baculovirus-mediated gene delivery (BacMam) enables convenient expression of GFP-Histone H3 fusion protein in the desired cell background. Coupled with a terbium (Tb)-labeled site-specific antibody, the assay allows identification of inhibitors of methyltransferases and demethylases acting on histone H3 lysine 4.

Physiologically Relevant Results

This assay allows investigation of histone H3 Lys4 trimethylation across various cellular backgrounds, including primary cells. It supports screening for inhibitors of methyltransferases and demethylases in physiologically relevant contexts.
The LanthaScreen™ Tb-anti-Histone H3K4me3 antibody shows higher affinity for trimethylated versus dimethylated lysine 4 peptides, though some cross-reactivity may occur. For further discrimination, the BacMam Histone H3K4me2 Cellular Assay Kit (Cat. No. A14164) can be used.

BacMam Convenience

Assays are fully homogeneous and addition-only, eliminating washing, lysate transfer, or separation steps required in Western blotting or ELISA.
Using the same BacMam GFP-Histone H3 reagent, other histone modifications (phosphorylation, methylation, acetylation) can be monitored by exchanging the Tb-labeled antibody. LanthaScreen™ TR-FRET technology provides reduced noise, minimal fluorescence interference, and high sensitivity, allowing assays with fewer cells than traditional methods.
