Target Information
NF-kB/Rel transcription factors are cytosolic in an inactive state, complexed with inhibitory IkB proteins. Activation of NF-kB typically occurs through phosphorylation-induced, proteasome-mediated degradation of IkB.
The key regulatory step involves activation of the IkB kinase (IKK) complex, composed of IKK alpha, IKK beta (catalytic subunits), and IKK gamma (regulatory subunit).
Phosphorylation at Ser177 and Ser181 (IKK beta) or Ser176 and Ser180 (IKK alpha) induces conformational changes leading to kinase activation.
