Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor in the immunoglobulin gene superfamily. The N-terminal region (aa1–107) is homologous to Ig kappa light chains, while the C-terminal extracellular region is proline-rich, forming a stalk for the Ig domain. CD7 is expressed on immature and mature T lymphocytes, T cell leukemias, NK cells, a subset of B cells, and malignant B cells. Cross-linking CD7 modulates T and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 interacts with phosphoinositol 3′-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of human CD34(+) CD38(-) cord blood cells expressing CD45RA, HLA-DR, and high CD7 levels can generate B cells, NK cells, and dendritic cells, but not myeloid or erythroid cells. The CD7(+) phenotype marks primitive human lymphoid progenitors.

CD7 may cooperate with CD28 during Treg function; mice deficient in both CD28 and CD7 show reduced Treg numbers and suppressive activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested in normal human peripheral blood cells; use 5 µL (0.125 µg)/test in 100 µL final volume (10⁵–10⁸ cells/test).
Light Sensitivity: Protect from light.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57).

Excitation: 488–561 nm
Emission: 695 nm
Laser Compatibility: Blue, Green, Yellow-Green lasers.

CD7 is directly associated with phosphoinositol 3′-kinase and is an important marker in leukemia diagnostics.
