Target Information
Receptor-tyrosine-kinase-like orphan receptor 1 (ROR1) is a tumor-associated surface protein predominantly expressed during embryogenesis, where it is involved in organ morphogenesis, nervous system development, and neural progenitor cell maintenance and survival. It is virtually absent from normal pediatric and adult tissues, except for low-level expression in immature B cell precursors (hematogones) and adipocytes.
ROR1 is notably overexpressed and considered a survival factor in several B lymphoid and epithelial malignancies, including chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL), acute lymphoblastic leukemia (ALL), marginal zone lymphoma, and lung adenocarcinoma.
Initially identified through PCR-based cloning in neuroblastoma cells, ROR1 was classified as an “orphan” receptor due to its unknown ligand. Wnt-5a has since been proposed as a candidate ligand, and ROR1 functions as a pseudokinase, promoting proliferation and resistance to apoptosis in cancer cells via Wnt-5a interaction and TCL1-mediated AKT co-activation.
ROR1 is expressed as a glycoprotein with extracellular immunoglobulin-like, Frizzled, and Kringle domains, and an intracellular tyrosine kinase domain.
