Product Specific Information

The B6H12 monoclonal antibody reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP and β3 integrins (CD51/CD61, CD41/CD61). It mediates bidirectional signaling regulating neural synaptic activity and macrophage phagocytosis. CD47 acts as a receptor for thrombospondin and can mediate activation or apoptosis in T cells.

This antibody has reported neutralizing activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for optimal results.

Blocking Buffer Usage:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample to antibody cocktails or single-color controls.

Technology:
NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimized spillover and spread.

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein interacting with integrins and regulating signaling pathways such as G protein activation. It plays roles in calcium signaling during cell adhesion, acts as a receptor for thrombospondin, and modulates signal transduction. CD47 interacts with CD172a (SIRPα) to inhibit macrophage phagocytosis and influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development. Expression is broad across tissues and reduced on Rh erythrocytes.
