Overview
The BD Rhapsody™ HT Xpress System utilizes BD’s proprietary, gentle, and robust micro-well based single-cell partitioning technology for high-throughput single-cell analysis.
It enables barcoding and capturing of more than 640,000 single cells per cartridge, making million-cell studies possible.
The system provides the same performance as the BD Rhapsody™ Express System but features a new flexible cartridge design that allows running up to 8 times the number of lanes simultaneously, supporting a broad range of cell inputs from 100–800,000 cells per cartridge.

The BD Rhapsody™ HT Xpress System provides enhanced cell capture, the ability to run million-cell studies easily, and comparable WTA sensitivity and gene correlation across all 8 cartridge lanes.
Cell Input and Capture Performance
A graphical and tabular representation of singlets and multiplets recovered across a range of targeted cell loads.
Human PBMCs were prepared and captured using the BD Rhapsody™ HT Single Cell Analysis System.
Data were collected from two 8-lane cartridges loaded separately by individual users on the same day for each of the different cell input loads, totaling six 8-lane cartridges for the entire range of cell input.
The number of cells sequenced may be less than the number captured due to bead loss, assay choice, and sample composition.
If sample viability is <100%, the number of viable cells captured may be lower than targeted.
Viable Cell Capture and Gene Correlation
Two cell types (Jurkat and Ramos) were pooled, stained with sample tags, and loaded at 55,000 cells per lane on an 8-lane cartridge versus a single-lane cartridge.

WTA Sensitivity and Comparison
20,000 cells per lane from a single PBMC donor were loaded into four lanes (A–D) in a BD Rhapsody™ 8-Lane Cartridge and processed on a BD Rhapsody™ HT Xpress System.
A second set of cells from the same donor was loaded into four lanes (E–H) of another cartridge and processed on a different BD Rhapsody™ HT Xpress System.
WTA libraries were prepared from subsampled beads at 6,000 cells.
All BD Rhapsody™ 8-Lane Cartridge results were compared to a BD Rhapsody™ Single-Lane Cartridge control.
