Target Information
Caspase 9 (ICE-like apoptotic protease 6, ICE LAP6, apoptotic protease Mch6, apoptotic protease activating factor 3, Apaf3)는 CARD 도메인을 포함하는 peptidase family C14의 구성원입니다. Caspase들은 시스테인 프로테아제로, 비활성 전구체 형태로 발현되어 특정 기질의 단백질 분해를 통해 세포사멸(apoptosis)을 매개합니다.
Caspase 9는 헤테로테트라머 형태로 활성화되며, 세포질에 존재하다가 활성화 시 미토콘드리아로 이동합니다. 활성화된 Caspase 9는 Caspase 3 및 Caspase 6을 절단·활성화시키며, Apaf-1과 결합하여 세포사멸 경로를 촉진합니다. 또한 DNA 손상에 의해 유도되는 apoptosis에서 ABL1/c-Abl 의존적 경로에 관여하며, poly(ADP-ribose) polymerase (PARP)를 절단합니다.
Caspase 9는 다양한 인간 조직에서 발현되며, 변이 또는 발현 저하는 뇌 발달 이상, 암, 알츠하이머병(AD) 등 신경퇴행성 질환과 관련이 있습니다.
