CD26 (dipeptidyl peptidase IV, DPP IV, adenosine deaminase (ADA) binding protein) is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family. It is expressed on lymphocytes and upregulated during T-cell activation, also found on activated B cells, NK cells, and epithelial cells. CD26 participates in T-cell activation, cell adhesion to extracellular matrix components such as fibronectin or collagen, and plays a role in HIV infection. It is identical to adenosine deaminase complexing protein-2 and the T-cell activation antigen CD26. As a membrane glycoprotein and serine exopeptidase, it cleaves X-proline dipeptides from the N-terminus of polypeptides. Alterations in CD26 activity are associated with malignant transformation, with enzymatic activity increasing with tumor grade. CD26 expression changes have also been reported in monocytes and CD4+ T cells in rheumatoid arthritis and systemic lupus erythematosus.
