Target Information
AP2S1 is a component of the adaptor protein complex 2 (AP-2). Adaptor protein complexes function in protein transport via vesicles in various membrane traffic pathways. They are vesicle coat components involved in cargo selection and vesicle formation.
AP-2 participates in clathrin-dependent endocytosis, where cargo proteins are incorporated into clathrin-coated vesicles (CCVs) destined for fusion with early endosomes. The clathrin lattice acts as a mechanical scaffold but does not directly bind membrane components.
Clathrin-associated adaptor protein (AP) complexes can bind both clathrin and the lipid/protein components of membranes, serving as major clathrin adaptors for CCV formation. AP-2 also acts as a cargo receptor to selectively sort membrane proteins involved in receptor-mediated endocytosis and plays a role in recycling synaptic vesicle membranes.
AP-2 recognizes Y-X-X-[FILMV] (Y-X-X-Phi) and [ED]-X-X-X-L-[LI] endocytosis signal motifs within cytosolic tails of transmembrane cargo molecules and may contribute to post-endocytic trafficking via the ARF6-regulated, non-clathrin pathway.
The AP-2 alpha and sigma subunits contribute to recognition of the [ED]-X-X-X-L-[LI] motif. AP-2 may also play a role in extracellular calcium homeostasis.
