Recommended positive control tissues include epithelial cells from pancreas, colon, or thyroid, though other tissues may also be suitable.
The primary antibody is designed for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissues for manual qualitative immunohistochemistry (IHC) analysis. It is intended for use after a primary tumor diagnosis has been made by conventional histopathology.
Cytokeratin 19 (CK19) is a type I acidic intermediate filament protein expressed in various human tissues except the liver. Since CK19 is absent in hepatocytes, this antibody is useful for identifying liver metastases. Clone BA17 reacts with CK19-positive MCF-7 cells and shows no reactivity with CK19-negative cells such as A431 or HeLa. In combination with HBME1, CK19 assists in differentiating thyroid carcinoma from benign thyroid follicles.
This antibody is optimized for use with formalin-fixed, paraffin-embedded sections. Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining enables visualization of antigens through sequential application of a primary antibody, a secondary antibody, an enzyme complex, and a chromogenic substrate. The resulting colorimetric reaction is observed under a light microscope for diagnostic interpretation.
A positive tissue control must accompany every staining procedure. Control tissues should contain both positive and negative staining regions to verify specificity and technique accuracy. External positive control materials should be freshly fixed, processed, and embedded similarly to patient samples. Weakly positive controls are preferred for detecting subtle changes in antibody sensitivity or reagent stability.
Negative control tissues (internal or external) should confirm the absence of specific staining and help assess background or non-specific binding. Any specific staining in negative controls invalidates the patient sample results.
Target Information
Cytokeratin 19 is a member of the intermediate filament protein family, showing significant biochemical diversity across human epithelial tissues. Cytokeratins range in molecular weight from 40–68 kDa with isoelectric points between 4.9–7.8 and are classified as types 1–20. Expression patterns vary by epithelial type and differentiation stage, making cytokeratin profiling valuable for distinguishing epithelial malignancies.
CK19 is not expressed in hepatocytes, making it a useful marker for identifying liver metastases. Cytokeratin antibodies are widely used in immunohistochemistry, cytopathology, and flow cytometry for tumor differentiation and epithelial cell characterization.
