Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, playing a central role in reproductive events associated with the establishment and maintenance of pregnancy.

PR exists as two forms:

• Form A (94 kDa)
• Form B (120 kDa)

These forms arise from alternative translation initiation sites. PR-B is transcriptionally active and regulates genes involved in endometrial maintenance, pregnancy, and inhibition of ovulation. PR-A differs by a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene cause reproductive abnormalities.
