Product Specific Information

  • This antibody is diluted and ready-to-use.
  • Recommended positive control tissue: Lymph Node (other tissues may also be suitable).
  • Intended for professional laboratory use in detecting TCL1 protein in formalin-fixed, paraffin-embedded tissue using manual qualitative IHC.
  • Used after primary tumor diagnosis by conventional histopathology.
  • TCL1A (p14TCL1) is involved in T-cell prolymphocytic leukemia (TPLL), which is associated with chromosomal rearrangements between TCRA locus (14q11) and TCL1A gene (14q32.13).
  • TCL1 is overexpressed in various lymphomas including Burkitt, AIDS-related non-Hodgkin, lymphoblastic, chronic lymphocytic, mantle cell, follicular, and diffuse large B-cell lymphoma.
  • Pretreatment by heat-induced or enzymatic epitope retrieval is recommended.
  • IHC staining visualizes antigens via sequential application of primary antibody, secondary antibody, enzyme complex, and chromogenic substrate with washing steps.
  • Results are interpreted microscopically to aid differential diagnosis.

Quality Control Guidance

  • A positive tissue control must be run with every staining procedure.
  • Control tissues should include both positive and negative staining areas.
  • External positive controls should be fresh specimens processed identically to patient samples.
  • Weakly positive tissues are preferred for detecting subtle reagent degradation.
  • Negative controls (internal or external) verify absence of non-specific staining and ensure validity of results.

Target Information

  • Overexpression of TCL1 gene is implicated in mature T-cell leukemia due to chromosomal rearrangements near TCR regulatory elements (Virgilio et al., 1998).
  • In normal T cells, TCL1 is expressed in CD4-/CD8- cells but not in later differentiation stages.
  • TCL1 acts as a coactivator of the cell survival kinase AKT (Laine et al., 2000).
