Target Information
Prolyl hydroxylase 4 (P4HTM)은 HIF-alpha 단백질을 변형시키는 프롤릴 하이드록실레이스 효소입니다. 전형적인 프롤릴 하이드록실레이스는 소포체 내에서 콜라겐을 변형시키지만, HIF는 세포 내 단백질로 콜라겐을 변형하는 효소와는 다른 부위를 인식합니다.
HIF는 산소 항상성 유지에 중요한 전사 복합체로, Prolyl hydroxylase는 세포가 산소를 감지하는 경로에서 핵심적인 역할을 합니다. 산소가 존재할 때, Prolyl hydroxylase는 HIF-alpha의 특정 프롤릴 잔기를 하이드록시프롤린으로 전환시켜 HIF-alpha의 분해를 유도합니다. 반대로 저산소 상태에서는 이 반응이 억제되어 HIF가 안정화되고 혈관신생이 증가합니다.
P4HTM은 HIF-alpha 단백질의 번역 후 4-hydroxyproline 형성을 촉매하며, 정상 산소 조건에서 세포 내 산소 센서로 기능할 수 있습니다. 또한 von Hippel-Lindau 유비퀴틴화 복합체를 통한 HIF의 프로테아좀 분해를 조절할 수 있습니다.
