Description:
The 9F10 monoclonal antibody reacts with human CD49d, the 150 kDa integrin alpha4 subunit. CD49d forms a complex with integrin beta1 (CD29), known as VLA-4, which serves as a receptor for fibronectin and VCAM-1 (CD106). This complex is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin beta7 and binds to MadCAM-1.
Applications Reported:
This antibody is reported for use in flow cytometric analysis.
Applications Tested:
Pre-diluted and validated by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.
Staining Panel Recommendation:
For panels with multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.
Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information
Integrin alpha 4 (CD49d, ITGA4) is part of the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta subunits. Alpha4 associates with beta1 or beta7 chains, forming receptors for ligands such as MadCAM, VCAM, and fibronectin. MadCAM is the principal ligand for integrin α4β7. HIV-1 envelope can mimic MadCAM by binding and signaling through integrin α4β7, the gut mucosal homing receptor for peripheral T cells.
