Target Information
VEGF (Vascular Endothelial Growth Factor)는 45 kDa의 동종이량체(disulfide-linked homodimeric) 당단백질로, 혈관신생(angiogenesis)에 관여하며 종양의 성장과 전이에 중요한 역할을 합니다. VEGF는 혈관 내피세포의 생존, 유사분열, 화학주성, 투과성 증가 등 다양한 생리적 효과를 유도합니다.
VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, PlGF가 포함되며, 이들은 주로 내피세포에 발현되는 티로신 키나아제 수용체 FLT1(VEGFR-1)과 FLK1/KDR(VEGFR-2)에 결합합니다. 이 신호전달 체계는 배아 혈관 발달에 필수적입니다.
또한 FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF에 높은 친화도로 결합하여 혈관신생을 억제하는 역할을 합니다. VEGF의 과발현은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes, Crow-Fukase syndrome)과 연관되어 있습니다.
